Molecular modeling studies of novel naphthyridine and isoquinoline derivatives as CDK8 inhibitors

Cell cycle is an important part of cellular activities. The selective inhibition of cyclin-dependent kinases (CDK) activity in tumor cells can lead to continuous cell proliferation. Thirty-nine CDK8 inhibitors were systematically investigated on the basis of a three-dimensional quantitative structure-activity relationship (3D-QSAR). Models for comparative molecular field analysis (q(2)=0.64,r(2)=0.98) and comparative molecular similarity index analysis (q(2)=0.609,r(2)=0.952) were obtained. Contour maps illustrated that bioactivity of inhibitors is most affected by steric, electrostatic, hydrogen bond donor, and receptor interactions of molecular groups. Twenty new CDK8 inhibitors (DS01-DS20) were designed based on the contour maps. The results of ADME prediction illustrated that the designed compounds had potential druggability. The binding mode between a ligand and receptor was explored through molecular docking and molecular dynamics. Results revealed that the hydrogen bond interaction with residue LYS52 remarkably affected the activity of these compounds. Further analysis indicated that the introduction of fluorine to an amino naphthyridine ring of compound 28 contributes to the improvement of molecular activities. Pharmacophore-based virtual screening and Surflex-Sim in the ZINC database of 1,30,000 molecules demonstrated that 14 compounds with an indazole ring might be antitumor inhibitors. 3D-QSAR, molecular docking, molecular dynamics and pharmacophore results are consistent. Repositioning HIV-1 Integrase Inhibitors for Cancer Therapeutics: 1,6-Naphthyridine-7-carboxamide as a Promising Scaffold with Drug-like Properties

Among a large number of HIV-1 integrase (IN) inhibitors, the 8-hydroxy-[1,6]naphthyridines (i.e., L-870,810) were one of the promising class of antiretroviral drugs developed by Merck Laboratories. In spite of its remarkable potency and efficacy, unfortunately upon completion of phase I clinical studies, development of L-870,810 was halted. Because of its desirable pharmacological and pharmaceutical properties we were intrigued to design novel analogues of L-870,810 with goals to (1) improve upon limitations of naphthyridine-7-carboxamides as antiviral agents and (2) to reposition their use as innovative cytotoxic agents for cancer therapeutics. Herein, we report on the design and synthesis of a series of 1,6-naphthyridine-7-carboxamides with various substitutions at the 5- and 8-positions. All the new 5-substituted-8-hydroxy-[1,6]naphthyridines were potent IN inhibitors and the 5-substituted-8-amino-[1,6]naphthyridines were significantly cytotoxic. Further optimization of the 5,8-disubstituted-[1,6]naphthyridines with structural variation on 7-carboxamide delivered novel compounds with significant cytotoxicity in a panel of cancer cell lines and effective inhibition against select oncogenic kinases.

The chelates display unique UV-light stability in solution and remain highly emissive after 100 min of strong UV irradiation (similar to 300 W.m(-2) at 345 nm). Moreover, they exhibit reversible luminescence intensity changes with varied pH values, and the response mechanism is investigated. Turn-on of the Eu(III) emission upon increasing pH is realized by ligand structure change from keto to enol anion form, resulting in red-shifted absorption band and suppressed quenching from solvents and N-H vibration upon deprotonating. The results show that these novel Eu(III) complexes are quite intriguing for potential application as bioimaging agents and pH probes.
