Various biologically relevant G-quadruplex DNA structures offer a platform for therapeutic intervention for altering the gene expression or by halting the function of proteins associated with telomeres. One of the prominent strategies to explore the therapeutic potential of quadruplex DNA structures is by stabilizing them with small molecule ligands. Here we report the synthesis of bisquinolinium and bispyridinium derivatives of 1,8-naphthyridine and their interaction with human telomeric DNA and promoter G-quadruplex forming DNAs. The interactions of ligands with quadruplex forming DNAs were studied by various biophysical, biochemical, and computational methods. Results indicated that bisquinolinium ligands bind tightly and selectively to quadruplex DNAs at low ligand concentration (similar to 0.2-0.4 mu M). Furthermore, thermal melting studies revealed that ligands imparted higher stabilization for quadruplex DNA (an increase in the T-m of up to 21 degrees C for human telomeric G-quadruplex DNA and >25 degrees C for promoter G-quadruplex DNAs) than duplex DNA (Delta T-m < 1.6 degrees C). Molecular dynamics simulations revealed that the end-stacking binding mode was favored for ligands with low binding free energy. Taken together, the results indicate that the naphthyridine-based ligands with quinolinium and pyridinium side chains form a promising class of quadruplex DNA stabilizing agents having high selectivity for quadruplex DNA structures over duplex DNA structures. A series of derivatives based on 10,10-dimethyl-5,10-dihydro-pyrido[4,3-b][1,6] naphthyridine-diphenylsulphone (DMDHNP-DPS) named 1a was designed with CH/ N and H/ CN substitution at the DPS acceptor unit to obtain blue thermally activated delayed fluorescence (TADF) materials. The parent molecule 1a was chosen from our previous report after CH/ N substitution at the DMAC donor fragment. The highest occupied molecular orbitals (HOMOs) and the lowest unoccupied molecular orbitals (LUMOs) were largely distributed over the DMDHNP donor and DPS acceptor units, respectively, resulting in a slight overlap between the HOMO-LUMO and hence a smaller singlet-triplet energy gap (DEST). Steric hindrance caused a large dihedral angle (E821-891) between the plane of the electrondonating DMDHNP unit and the electron-accepting DPS unit in the substituted derivatives. Calculated results indicated that the DEST values of H/ CN substituted derivatives were smaller than those of the corresponding CH/ N derivatives which were favorable for the reverse intersystem crossing (RISC) process from the lowest excited triplet states (T1) to the lowest excited singlet (S1) states and ultimately to the ground state (S0) causing delayed emission. The emission wavelengths (lem) of all the designed molecules were found to be in the range of 397-497 nm. The incorporation of the -NQatom or the -CN group at the ortho and meta positions of DPA reduced the transition energies from LUMO -HOMO in the S1 states, resulting in a red-shift. Moreover, the lem values displayed a more substantial bathochromic-shift as the number of -NQatoms or -CN groups increased. The two of the designed molecules (1h and 1i) showed sky-blue emission (494 nm and 497 nm), and the four of the investigated compounds (1c, 1d, 1f, and 1g) displayed blue emission (416 nm, 447 nm, 437 nm, and 432 nm, respectively) indicating that these investigated derivatives were efficient sky-blue to blue TADF candidates. Among all the investigated derivatives, the smaller DEST values for the designed systems 1f (0.03 eV) and 1g (0.02 eV) and appropriate lem values of 437 nm and 432 nm make them excellent candidates for blue TADF materials. Our theoretical investigation might offer hints for the construction of efficient blue TADF-based organic light emitting diodes (OLEDs) in the future.

In this work, we describe the synthesis of 1,8,10-trisubstituted anthracenyl scaffolds that, bearing boronic acid functionalities, can act as multiple H-bonding donor systems. The trisubstituted anthracenyl derivatives are synthesized following two main synthetic pathways. Whereas in the first approach trisubstituted anthracenyl derivatives are prepared through the regioselective addition of the relevant organomagnesium nucleophile to 1,8-dichloroanthraquinone, in the second avenue a triflate-bearing anthracene is prepared by reduction of the anthraquinone into the anthrone precursor and functionalized through metal-catalysed cross-coupling reactions. Complementary studies of the Na2S2O4-mediated reduction of 1,8-dichloroanthraquinone allowed to shed further light on the possible mechanism of formation of the anthrone precursor, suggesting the presence of a cis-diol intermediate undergoing antiperiplanar elimination. Solid-state X-ray diffraction investigations of the bisboronic acids show that the molecules self-assemble into dimers through the formation of four H-1-bonds established between the anti-syn conformers of the boronic acid moieties. H-1-NMR titrations between bisboronic acids and tetra H-bond acceptor, diisoquinolino-naphthyridine, showed a significant shift of the -B(OH)(2) proton resonances, suggesting the presence of H-bonding interactions between both molecules. The new pyrrolo[3,4-c]pyridines and 2,7-naphthyridine derivatives have been synthesized. 4-Hydroxy-8-methyl-l-oxo-6-phenyl-1,2-dihydro-2,7-naphthyridine-3-carboxylic acid hydrazide (4) was the key intermediate for the synthesis of the novel derivatives of various chemical structures: Schiff bases, 1,3,4-oxadiazoles, pyrazoles, carbohydrazides, semi- and thiosemicarbazides. A novel series of heterocyclic systems fused coumarins and chromones were efficiently synthesized from chemical transformations of 6,8-dimethylchromone-3-carbonitrile (1) with a variety of carbon nucleophilic reagents.
