Step 1. Ethyl 1- (2, 4-difluorophenyl)-6-fluoro-4-oxo-7- (piperazin-1-yl)-1, 4-dihydronaphthyridine-3- carboxylate: Ethyl 7-chloro-1- (2, 4-difluorophenyl)-6-fluoro-4-oxo-1, 4-dihydronaphthyridine- 3-carboxylate (766 mg, 2.0 mmol) and piperazine (430 mg, 5. 0 mmol, 2.5 equiv) were dissolved in 20 mL of pyridine and allowed to stir at room temperature for 24 h. The solvent was removed in vacuo, and the crude product was taken up in CH2C12, washed with 5% aq Na2CO3, then water. The organic layer was dried (Na2SO4), evaporated in vacuo, and the resulting white solid was purified on silica gel column using a linear gradient (100% CH2C12 to 10% MeOH in CH2C12) to give a white solid.
