A mixture of 7-chloro-1 ,2-dihydro-1 ,8-naphthyridin-2-one (1 g, 5.53 mmol), 3-bromo-1 , 1- dimethoxypropane (1.1 g, 6.09 mmol) and K2CO3 (1.1 g, 8.31 mmol) in DMF (20 mL) was heated at 70C for 4 h. The mixture was then allowed to cool to room temperature, poured into H2O (30 mL) and extracted with EtOAc (30 mL x 3). The combined organic extracts were washed with H2O (50 mL x 2), brine (50 mL), dried over Na2S04, filtered and concentrated under reduced pressure. The residue was purified by silica gel chromatography using 30% EtOAc/petroleum ether to give a yellow solid of 7-chloro-1-(3,3- dimethoxypropyl)-1 ,2-dihydro-1 ,8-naphthyridin-2-one 6a (1 g, 64%).
